Figure 5 BTV induces mitochondria-mediated apoptosis in renal cancer cells. GAPDH and β-actin were used as loading control for cytosolic fraction and COX IV was used for mitochondrial fraction. (A). Cells were treated as described previously. Detection of P53, Bax expression. The p53 and Bax protein expression levels were upregulated in cancer cells but not in HPTEC. (B) The cytochrome c protein level was elevated in the cytosolic fractions in cancer cells. (C) Detection of the activated form of caspase-3, -9, and cleaved poly (ADP-ribose) polymerase (PARP). Pre-treatment of CAKI-1 and OS-RC-2 cells with Z-VAD-FMK (10 μM) for 24 h before BTV infection (MOI=10) markedly inhibited the expression of the activated form of caspase-3 and PARP.
