Figure 3 mRNA expression levels of ATM, NFKB1, SOD2, INSR, and TIGAR in visceral preadipocytes (N, H), differentiated (NN, NH, HN, HH), and mature (NNN, NNH, NHN, HNN, HHH) adipocytes in normoglycemic (N, 5.5mM glucose) and hyperglycemic (H, 30 mM) milieu. Data are presented as mean±SD. Green circles mark merely normoglycemic variants (N, NN, NNN) while red squares denote overly hyperglycemic ones (H, HH, HHH) during the process of visceral adipogenesis (tested with ANOVA with post hoc Tukey’s test and marked with green and red asterisks, respectively). Black triangles denote either differentiated or mature adipocytes with single-stage hyperglycemic exposure (NH, HN, NNH, NHN, HNN). Black asterisks were used to mark significant impact of hyperglycemia measured at the stage of preadipocytes (H vs N, by two-tailed t test), differentiated and mature adipocytes (NH, HN, HH vs NN and NNH, NHN, HNN, HHH vs NNN by one-way ANOVA with post hoc Dunnett’s test). Black asterisks were also used to indicate changes occurring during normoglycemic differentiation and maturation of hyperglycemia-treated preadipocytes (H vs HN vs HNN, tested with ANOVA with post hoc Tukey’s test). Conjunct lines below the asterisks denote all significant differences between the first indicated variant on the left and subsequent variants indicated by particular segments. Significant differences are indicated accordingly to the criteria: * 0.05≥P>0.01, ** 0.01≥P>0.001, *** P≤0.001, based on −ΔCt values. The figure was generated using GraphPad Prism 6.0 (GraphPad Software Inc.).
